Efficacy of a 12-Week Simeprevir Plus Peginterferon/Ribavirin (PR) Regimen in Treatment-Naïve Patients with Hepatitis C Virus (HCV) Genotype 4 (GT4) …
2017
Background HCV GT4 accounts for up to 20% of HCV infections worldwide. Simeprevir, given for 12 weeks as part of a 24- or 48-week combination regimen with PR is approved for the treatment of chronic HCV GT4 infection. Primary study objectives were assessment of efficacy and safety of simeprevir plus PR in treatment-naïve patients with HCV GT4 treated for 12 weeks. Primary efficacy outcome was sustained virologic response 12 weeks post-treatment (SVR12). Additional objectives included investigation of potential associations of rapid virologic response and baseline factors with SVR12. Methods This multicentre, open-label, single-arm study (NCT01846832) evaluated efficacy and safety of simepre…
Nodular Lesion in Cheek With a Hair Tuft
2017
Trichostasis spinulosa is a rarely diagnosed disorder of the pilosebaceous unit, characterized by retention of telogen hairs within the dilated follicles. A hair tuft can be seen protruding from the follicles. We present a case of trichostasis spinulosa associated with intradermal melanocytic nevi, where dermoscopy helps to identify this entity.

Efficient selection of silenced primary cells by flow cytometry
2007
Background: RNA interference has emerged as a new and potent tool to knockdown the expression of target genes and to investigate their functions. For short time experiments with mammalian cell lines, RNA interference is typically induced by transfecting small interfering RNAs (siRNAs). Primary cells constitute important experimental systems in many studies because of their similarity to their in vivo counterparts; however, transfection of these cells has been found to be difficult. As a consequence, RNA interference of primary cells may result in mixed phenotypes because of the simultaneous presence in the same preparation of transfected and nontransfected cells. This may be particularly in…
Flow cytometric kinetic assay of the activity of Na+/H+ antiporter in mammalian cells.
2004
Background The Na+/H+ exchanger (NHE) of mammalian cells is an integral membrane protein that extrudes H+ ion in exchange for extracellular Na+ and plays a crucial role in the regulation of intracellular pH (pHi). Thus, when pHi is lowered, NHE extrudes protons at a rate depending of pHi that can be expressed as pH units/s. Methods To abolish the activity of other cellular pH-restoring systems, cells were incubated in bicarbonate-free Dulbecco's modified Eagle's medium buffered with HEPES. Flow cytometry was used to determine pHi with 2′,7′-bis-(2-carboxyethyl)-5-(and-6)-carboxyfluorescein acetoxymethyl ester or 5-(and-6)-carboxy SNARF-1 acetoxymethyl ester acetate, and the appropriate fluo…
Near Infrared Spectroscopy Detection and Quantification of Herbal Medicines Adulterated with Sibutramine.
2015
There is an increasing demand for herbal medicines in weight loss treatment. Some synthetic chemicals, such as sibutramine (SB), have been detected as adulterants in herbal formulations. In this study, two strategies using near infrared (NIR) spectroscopy have been developed to evaluate potential adulteration of herbal medicines with SB: a qualitative screening approach and a quantitative methodology based on multivariate calibration. Samples were composed by products commercialized as herbal medicines, as well as by laboratory adulterated samples. Spectra were obtained in the range of 14,000-4000 per cm. Using PLS-DA, a correct classification of 100% was achieved for the external validatio…
Methomyl analogues with increased biological activity towards F7T maize mitochondria
1987
Abstract Methomyl analogues were synthesized by substituting alkyl moieties (C 2 -C 21 ) in the place of the carbamic methyl. They were assayed on mitochondria isolated from male sterile (F 7 T) and male fertile (F 7 N) maize. They had no action on F 7 N mitochondria. The heptadecyl (C 17 ) and heneicosanyl (C 21 ) derivatives had no conspicuous effect on F 7 T mitochondria. By contrast, the ethyl, propyl, butyl, nonyl, tridecyl (C 13 ) and pentadecyl (C 15 ) derivatives had the same type of activity as Methomyl on F 7 T mitochondria, namely stimulation of NADH oxidation and inhibition of malate oxidation. Moreover, the concentration at which they were maximally effective decreased from 10 …
Combined Anchoring: Prosecution and defense claims as sequential anchors in the courtroom
2021
Purpose When making judgements under uncertainty not only lay people but also professional judges often rely on heuristics like a numerical anchor (e.g., a numerical sentencing demand) to generate a numerical response. As the prosecution has the privilege to present its demand first, some scholars have speculated about an anchoringbased unfair disadvantage for the defence (who has the last albeit less effective word in court). Despite the plausibility of this reasoning, it is based on a hitherto untested assumption that the first of two sequential anchors exerts a greater influence on a later judgement (a primacy effect). We argue that it is also conceivable that the last word in court h…
Using the fibre structure of paper to determine authenticity of the documents: analysis of transmitted light images of stamps and banknotes.
2014
A novel method is presented for distinguishing postal stamp forgeries and counterfeit banknotes from genuine samples. The method is based on analyzing differences in paper fibre networks. The main tool is a curvelet-based algorithm for measuring overall fibre orientation distribution and quantifying anisotropy. Using a couple of more appropriate parameters makes it possible to distinguish forgeries from genuine originals as concentrated point clouds in two- or three-dimensional parameter space.
GHB related acids are useful in routine casework of suspected GHB intoxication cases
2021
Abstract GHB related acids (3,4-dihydroxy butyric acid, 2,4-dihydroxy butyric acid and glycolic acid) are produced through oxidative GHB metabolism. These analytes could be potential biomarkers to ensure the diagnosis of a GHB intoxication and even prolong the detection window. Within this study, forensic routine cases were measured to consider the potential of additional gas chromatographic mass spectrometric analysis on these acids. 17 GHB positive real cases (10 serum samples and 7 urine samples) and 40 cases with suspicion of drugging in DFC cases and negative GHB results (21 serum samples and 19 urine samples) were evaluated. Increased GHB related acid concentrations were detected in a…
